摘要:
Metastasis is a leading cause of death for gastric cancer (GC) patients. However, little is known about the mechanism underlying GC metastasis. Here, we found a new lncRNA GMAN (gastric cancer metastasis associated long non-coding RNA) that is upregulated in GC tissues and correlated with metastasis and poor prognosis. GMAN is a sense lncRNA that overlaps with Ephrin-A1 and enhances GC cell metastasis by facilitating Ephrin-A1 mRNA translation. GMAN promotes Ephrin-A1 mRNA translation by competitively binding to a previously unrecognized GMAN antisense RNA (GMAN-AS) whose sequence is complementary to both GMAN and Ephrin-A1. Targeting of GMAN by the CRISPR/Cas9 system robustly reduces Ephrin-A1 protein levels and eliminates GC cell metastasis. Moreover, an in vivo CRISPR/Cas9-based treatment targeting GMAN significantly attenuated GC cell metastasis. These results reveal a hitherto uncharacterized mechanism of cancer metastasis by a triple RNA competition for translational regulation, and indicate a potential therapeutic approach to control GC metastasis.
