Polycomb Group (PcG) proteins regulate plant developmental stage transitions through the repression of target gene transcription. PcG proteins function by forming complexes, such as Polycomb Repressive Complex (PRC) 1 and 2, which are responsible for the deposition of chromatin marks (H3K27me3 and H2AK121ub1) associated with gene repression. These PcG dependent histone modifications undergo dynamic changes throughout the plant life cycle. However, both the relationship between H3K27me3 and H2AK121ub1 and the mechanisms behind the genome wide establishment and maintenance of these marks by PcG proteins remain elusive. Here, I provide evidence to answer these questions. Firstly, I demonstrated that H2AK121ub1 can be targeted to genes independently of PRC2 activity and H3K27me3 localization. Secondly, I showed that telobox elements and TELOMERE REPEAT BINDING proteins (TRBs) work as genome-wide recruiters of PRC2 for H3K27me3 establishment. Finally, I described the involvement of Enhancer Of LHP1 (EOL1) in H3K27me3 maintenance through the interaction of PcG proteins and DNA replication machinery during cell division. Through studying the molecular mechanisms of PcG mediated gene repression, my research has contributed to a better understanding of the complexity of gene transcription control and plant developmental regulation.
