题目:Structure and function of enzymes involved in phosphorylation and adenylylation

Reversible phosphorylation, executed by kinases and phosphatases, regulates almost every aspect of cellular life. A lesser-known but highly analogous process is adenylylation, in which the adenosine monophosphate (AMP) group of ATP is attached to and removed from other molecules to control their function. My research concerns the fundamentals of above processes, aiming at elucidating the molecular mechanism of enzymes that catalyze these reactions. I will discuss two such enzymes: Fic from the bacterial pathogen Histophilussomni, which adenylylatesan tyrosine residue of the Rho-family GTPases thereby collapsing the host cell actin cytoskeleton; and PTPMT1, a mitochondrial phosphatase that is required for cardiolipin biosynthesis in mammalian cells.
