Single-cell genetic approach was used to study subunit composition of synaptic AMPA receptors, and an innovative molecular replacement approach was employed to examine novel mechanisms underlying the regulation of synaptic strength in the hippocampus. In addition, I will discuss work-in-progress in which glutamatergic synaptic transmission is silenced in individual pyramidal neurons in the hippocampus in a mosaic fashion in vivo, which provides unique approaches to study the cell-autonomous role of excitatory synaptic transmission in the regulation of neuronal development and function. At the end of the talk, future research plans will also be discussed.
