During tissue formation, cells face two major challenges. First, progenitor cells need to maintain a balance between self-renewal and differentiation to ensure the generation of the correct number of different cell types. Second, differentiated cells need to maintain appropriate organization to confer organ functionality. I have utilized zebrafish as a model system to understand the regulation of these processes in vivo. First, I will describe how neural progenitors in the spinal cord switch from progenitor states to differentiated states. Combining genetic manipulation, in vivo imaging and a novel method to study cell signaling in live embryos, I discovered that Notch signaling regulates Hedgehog (Hh) responsiveness of neural progenitors and that the attenuation of both Notch and Hh signaling is required for the transition from the progenitor state to the differentiated state. Second, I will describe a novel function of primary cilia, small cellular protrusions, in the maintenance of muscle integrity. I discovered that genetic ablation of cilia results in defects in muscle attachments. I will describe how cilia and Hh signaling regulate muscle integrity and discuss how these findings inform strategies to promote tissue repair.
